Abstract

The rapid maturation of the field of biopharmaceutical formulation is the result of the simultaneous development of a thermodynamic mechanism for protein-solvent interaction and identification of natural chemicals employed by nature to stabilize proteins in response to environmental stresses. In general, these cosolvents are excluded from the protein surface. Proteins are maintained in their native folded conformations by these cosolvents as a result of the highly unfavorable interaction between cosolvents and peptide backbones, which would be exposed to the cosolvent upon unfolding.
